5-Bromo-DMT is a psychedelic tryptamine and brominated indole alkaloid related to DMT. It is the 5-bromo derivative of DMT and is classified as both psychedelic and sedative. The compound occurs naturally in several marine invertebrates, including the sponges Smenospongia aurea, Smenospongia echina, and Verongula rigida. Animal studies have suggested possible sedative or antidepressant applications, but these uses remain tentative.1

Pharmacodynamics

5-Bromo-DMT acts primarily as a partial agonist at the serotonin 5-HT2A receptor, with reported efficacy ranging from 34% to 100%. It also acts as a weak full agonist at 5-HT1A and shows affinity for 5-HT2B, 5-HT2C, and the serotonin transporter, where it is a very weak serotonin reuptake inhibitor. In rodents, it failed to significantly produce the 5-HT2A-linked head-twitch response and antagonized the response induced by 5-fluoro-DMT2, while producing antidepressant-like, sedative-like, hypothermic, and psychoplastogenic effects.
